Correlation between rs4129267 Polymorphism on IL-6R Gene and Pediatric Asthma Susceptibility and Serum IgE Level of Han Chinese Children

Abstract

Abstract: Background　Research has confirmed the significant correlation between rs4129267 polymorphism on IL-6R gene and pediatric asthma suseptibility,but currently,the distribution of rs4129267 polymorphism among Han Chinese children and its correlation with serum IgE level have not been studied.Objective　To investigate the correlation between rs4129267 polymorphism on IL-6R gene and pediatric asthma susceptibility and serum IgE level of Han Chinese children.Methods　410 asthma children from Department of Pediatrics of Guangdong Women and Children Hospital were selected as asthma group from 2013 to 2014.513 non asthma children in the same hospital were selected as control group during the same period.The DNA samples of two groups were extracted.Then the sites of rs4129267 and rs527236128 at IL-6R gene were genotyped by SNaPshot method.The total serum IgE level of two groups was determined by ELISA.Results　The genetic distributions of rs4129267 and rs527236128 were accorded with Hardy-Weinberg equilibrium （χ2=3.264,2.135,P>0.05）.The genotype frequency and allele frequency of rs4129267 were significantly different between the two groups （P<0.05）.The genotype frequency and allele frequency of SNP rs527236128 were not significantly different between the two groups （P>0.05）.The Logistic regression result showed that TT genotype was the risk factor to pediatric asthma （P<0.05）.The total serum IgE level of control group was lower than that of asthma group （P<0.05）.There was no significant difference in the total serum IgE level among different genotypes in two groups （P>0.05）.The total serum IgE levels at all kinds of genotypes of control group were lower than those of asthma group （P<0.05）.Conclusion　rs4129267 polymorphisms on IL-6R gene is related to pediatric asthma suseptibility in Han Chinese children,but is not related to their serum IgE levels.

摘要： 背景　有研究证实，白介素6受体（IL-6R）基因中的rs4129267位点多态性与哮喘易患性显著相关，但尚无评估rs4129267位点多态性在汉族儿童中分布情况及其与血清IgE水平相关性的研究。目的　研究IL-6R基因rs4129267位点多态性与汉族儿童哮喘易患性及血清IgE水平的关系。方法　选取2013—2014年于广东省妇幼保健院儿科就诊的哮喘患儿410例为哮喘组，同期选取广东省妇幼保健院住院的非哮喘患儿513例为对照组。提取两组患儿基因组DNA，采用SNaPshot法对IL-6R基因rs4129267、rs527236128位点进行基因分型，采用酶联免疫吸附试验（ELISA）法检测两组患儿血清总IgE水平。结果　两组患儿rs4129267位点和rs527236128位点的基因型分布均符合Hardy-Weinberg平衡（χ2=3.264、2.135，P>0.05）。两组患儿rs4129267位点基因型频率和等位基因频率比较，差异有统计学意义（P<0.05）。两组患儿rs527236128位点基因型频率和等位基因频率比较，差异无统计学意义（P>0.05）。Logistic回归分析结果显示，TT基因型是儿童哮喘的影响因素（P<0.05）。对照组患儿血清总IgE水平低于哮喘组（P<0.05）。对照组、哮喘组组内不同基因型患儿血清总IgE水平比较，差异无统计学意义（P>0.05）；对照组各基因型患儿血清总IgE水平均低于哮喘组（P<0.05）。结论　IL-6R基因rs4129267位点多态性与汉族儿童哮喘易患性有关，而与哮喘患儿血清IgE水平无关。
