钙激活氯通道TMEM16A在大鼠心房和心室中的表达差异

Abstract: Objective To explore the expression of calcium activated chloride channel (CaCCs) TMEM16A in rat atrial and ventricular tissues,so as to provide a new therapeutic target for clinical treatment of myocardial ischemia,arrhythmia and heart failure.Methods From October 2014 to September 2015,18 clean grade Sprague-Dawley rats were given 1 week of adaptive feeding,then,the heart was taken out after intraperitoneal injection of 10% chloral hydrate,and the atrial and ventricular tissues were isolated.The expression of TMEM16A and its mRNA in atrial and ventricular tissues were detected by Western blotting methods and real-time fluorescence quantitative reverse transcription PCR (RT-qPCR),respectively.Results The expression levels of TMEM16A in the rat atrial tissues were higher than those in the ventricular tissues〔(0.46±0.02) vs. (0.33±0.03),P<0.01〕.The expression levels of TMEM16A mRNA in the rat atrial tissues were higher than those in the ventricular tissues〔(3.55±0.17) vs. (1.00±0.03),P<0.01〕.Conclusion TMEM16A CaCCs was expressed in both atrial and ventricular tissues of rats,and the expression level in the atrial tissue was higher than that in the ventricular tissue.

摘要: 目的 探究钙激活氯通道(CaCCs)TMEM16A在大鼠心房和心室组织中的表达情况,为临床治疗心肌缺血、心律失常以及心力衰竭等疾病提供新的治疗靶点。方法 2014年10月—2015年9月,将18只清洁级Sprague-Dawley大鼠适应性饲养1周,采用10%水合氯醛溶液腹腔注射麻醉后取心脏,分离心房和心室组织进行实验。采用Western blotting法和实时荧光定量反转录PCR(RT-qPCR)法分别检测大鼠心房和心室组织中TMEM16A及其mRNA水平。结果 大鼠心房和心室组织TMEM16A水平分别为(0.46±0.02)和(0.33±0.03),心房和心室组织TMEM16A mRNA水平分别为(3.55±0.17)和(1.00±0.03),大鼠心房组织中TMEM16A及其mRNA水平高于心室组织(P<0.01)。结论 CaCCs TMEM16A在大鼠心房和心室组织中均有表达,其在心房组织中的水平高于心室组织。
